Co-authored with Carson Key, Kristen Froehlich, and Thom LaBean

This chapter outlines detailed protocols for designing, producing, and evaluating RNA origami-based anticoagulants that use thrombin RNA aptamers. These programmable RNA nanostructures show promising therapeutic potential with high specificity, reversible activity, and improved pharmacokinetics — foundational to the RNA origami platform we're building at Helixomer.

šŸ”¬ We explore how self-folding, single-stranded RNA origami can deliver superior anticoagulant effects compared to free aptamers or DNA nanostructures, including using 2′F-modified nucleotides, heat-annealing protocols, and aPTT assays for activity testing.
