Updated rabbit data confirm that Hex01, Helixomer’s IV anticoagulant, remains active in the bloodstream for 2 to 3.5 hours depending on dose — long enough to cover a typical surgery from a single dose.

This is a significant result for the aptamer field. Most aptamer-based drugs are cleared from the body within minutes, which has historically kept this class out of the clinic. Prior industry attempts to extend half-life relied on polyethylene glycol (PEG) as a bulking agent. PEG is highly immunogenic: anti-PEG antibodies are present in up to 70% of the general population, and anaphylactic reactions have paused or terminated competitor programs.

Hex01 achieves its half-life through two design choices: a chemical modification that resists enzymatic degradation, and an RNA Origami 3D structure large enough to evade rapid kidney clearance — without PEG. No immune liability.

Hex02, the matched reversal agent, switches off anticoagulation immediately on demand. Hex01 is also shelf-stable at room temperature for more than a year in solution.
